Noninvasive Fetal RHD Genotyping Using Cell-Free Fetal DNA
Measurement of cell-free DNA for fetal genotyping for RhD antigen may be medically necessary when all of the following criteria are met: the pregnancy may be at risk for alloimmunization due to maternal RhD negative status or maternal red cell antigen antibodies, paternal antigen typing is unavailable or heterozygous, and amniocentesis is declined or contraindicated. The policy history records a 10/15/2025 update that changed CPT codes 81403 and 81479 from investigational to medically necessary. Code 0632U was added to the code reference effective 07/01/2026 as a new CPT code for multiplex PCR and next-generation sequencing of circulating cell-free DNA from plasma of pregnant individuals known to be RhD negative, reported as detected or not detected.
